Um die enorme Komplexität, derer sich die Natur in diesen Signalwegen bedient, möglich zu machen bzw. auch aufrecht zu erhalten und pathologische Fehlregulationen zu verhindern, muss auch der zugrunde liegende Proteaseapparat entsprechend komplex reguliert werden. Die Cysteinprotease Legumain ist ein Paradebeispiel dafür, wie facettenreich diese Regulation sein kann. Legumain wurde ursprünglich in Endo-Lysosomalen Zellkompartimenten entdeckt, in denen es als Schlüsselenzym der Prozessierung von Fremdantigenen für die anschließende Präsentation durch Immunrezeptoren gilt. Ein Zuwenig oder Zuviel an Legumain ist mit schwerwiegenden Erkrankungen wie Multiple Sklerose und Krebs assoziiert. Legumain ist in Zellkompartimenten mit saurem pH (Lysosomen) aktiv, wird jedoch irreversibel inaktiviert bei neutralem pH, also theoretisch im extrazellulären Milieu. Bis zum jetzigen Zeitpunkt waren grundlegende Prinzipien, etwa wie die Aktivierung von der Proform zum aktiven Enzym erfolgt, wie die Proteaseaktivität reguliert wird und wie sich unterschiedliche zelluläre Lokalisierungen erklären lassen, noch weitgehend unverstanden. In dieser Arbeit beschreiben wir die grundlegende biochemische und strukturbiologische Charakterisierung von Legumain.<br /> eng: Biological signaling involves a complex network of decision making and transferring molecules that must be regulated by proteases, key players in health and disease. The cysteine protease legumain exemplifies how intricate multifactorial processes combine in zymogen activation and activity regulation. These principles are key to accomplish its demonstrated roles in (auto-)immunity and cancer.<br />However, the exact mechanism of the activation process and the resulting consequences on legumain activity and localization remained unclear. The aim of the here presented work was to study biochemical and structural properties of human (pro)legumain to get an in-depth understanding of its enigmatic autocatalytic activation and activity regulation principles.
