Sechs Publikationen wurden als Erstautor verfasst, wobei vier davon in international begutachteten Journalen publiziert wurden und zwei sich derzeit unter Begutachtung befinden. Zusätzlich wurden weitere drei Publikationen als Co-Autor mitverfasst, die sich mit Trenntechniken und schwingungsspektroskopischen Analysen auseinandersetzen. Eine dieser Arbeiten wurde bereits publiziert, eine befindet sich derzeit unter Begutachtung und eine weitere liegt als Manuskriptentwurf vor.<br />Die erste Publikation (Teil B, Kapitel 2) beschreibt die Anwendung statistischer Versuchsplanung für die Methodenentwicklung und -optimierung in der Probenvorbereitung. Ein selbst hergestelltes Polymer wurde für die Aufreinigung und Isolierung von Rosmarinsäure aus einem Rosmarinextrakt verwendet. Ein voll-faktorielles Design wurde eingesetzt, um die Waschbedingungen und die Analyt-Wiederfindungen zu optimieren. Weiters konnte eine selbst entwickelte „Gaussian peak fit“-Funktion eingesetzt werden, um die Aufreinigungseffizient in Chromatogrammen objektiv quantifizieren zu können.<br />In der zweiten Publikation (Teil B, Kapitel 3) konnte ein neuartiges Polymer, basierend auf einer Co-Polymerisation von Vinylbenzylchlorid und Ethylenglycoldimethacrylat, entwickelt werden. In weiterer Folge wurde dieses mit vier stickstoffhaltigen Verbindungen funktionalisiert. Nach einer ausführlichen Charakterisierung wurde ein Test zur Anreicherung von ausgewählten Pharmazeutika und Estrogenen durchgeführt. Auch die Anwendung auf Wasserproben zeigte gute Ergebnisse in allen Experimenten.<br />Die dritte Publikation (Teil B, Kapitel 4) beschreibt die Synthese eines selbst hergestellten Co-Polymers zur selektiven Isolierung von Koffein aus Schwarz- und Grüntee Proben. Hierzu wurde Glycidylmethacrylat und Ethylenglycoldimethacrylat polymerisiert und anschließend mit Histidin modifiziert. Weiters wurden Untersuchungen der maximalen Adsorptionskapazität und der Adsorptionskinetiken durchgeführt. Das Material zeigte eine exzellente Selektivität für die Isolierung von Koffein aus Teeproben im Vergleich zu kommerziell erhältlichen Materialien.<br />Die vierte Publikation (Teil B, Kapitel 5) beschäftigt sich mit der Methodenentwicklung einer hochleistungsflüssigchromatographischen Methode zur Quantifizierung von polyzyklischen aromatischen Kohlenwasserstoffen. Verschiedene Festphasenextraktionsmaterialien wurden verglichen, um diese Komponenten in Pflanzenextrakten ausreichend bestimmen zu können. Dabei wurde auch ein selbst hergestelltes Polymer basierend auf der Synthese der zweiten Publikation (Teil B, Kapitel 3) verwendet. Die Methode wurde durch Variation der Elutionsmittel optimiert und konnte anschließend erfolgreich validiert werden.<br />In der fünften Publikation (Teil B, Kapitel 6) wird die Synthese neuer ionischer Flüssigkeiten sowie deren Anwendung in der Flüssigphasenmikroextraktion beschrieben. Nach einer Charakterisierung mittels Kernspinresonanzspektroskopie wurden mehrere Extraktionsexperimente durchgeführt, um die Eigenschaften der Extraktionssysteme zu ermitteln. Wiederfindungen, Anreicherungsfaktoren und Extraktionszeiten konnten dabei in wässrigen Proben, die Pharmazeutika und Estrogene enthielten, optimiert werden.<br />Die sechste Publikation (Teil B, Kapitel 7) beschreibt die Entwicklung eines molekular geprägten Polymersystems zur selektiven Isolierung von Acetylsalicylsäure aus wässrigen Lösungen. Eine ausführliche Charakterisierung sowie mehrere Adsorptionsexperimente wurden dabei durchgeführt. Dabei konnten maximale Adsorptionskapazitäten, Adsorptionszeiten sowie Selektivitäten festgestellt werden. Eine Anwendung für die Abwasseraufbereitung wurde durch Isolations- und Regenerationsstudien überprüft. | ||
520 | |a eng: Focus of this cumulative dissertation was set on the development of new materials and methods for isolation and determination of molecules with toxicological and pharmacological relevance. Six publications were written as first author, whereby four of them are published and two publications are submitted to peer-reviewed journals. Additionally, three research articles dealing with separation science and vibrational spectroscopy were co-authored (one published, one submitted and one manuscript draft in preparation).<br />The first publication (part B, chapter 2) describes the application of design of experiments for method development and optimization in sample preparation using solid-phase extraction (SPE). A self-synthesized mixed-mode polymer was used for clean-up and isolation of rosmarinic acid from rosemary extracts. A full-factorial design was implemented to optimize wash conditions and recoveries. In parallel, a Gaussian peak fit function was developed for objective quantification of clean-up efficiency in chromatograms after sample preparation.<br />In the second publication (part B, chapter 3) a novel polymeric sorbent based on co-polymerization of vinylbenzyl chloride and ethylene glycol dimethacrylate was synthesized and functionalized with four different nitrogen-containing compounds by nucleophilic substitution. After comprehensive characterization the synthesized mixed-mode materials were tested as SPE sorbents for enrichment of selected pharmaceuticals and endocrine-disrupting compounds. Additionally, application was also tested on different environmental water samples.<br />The third publication (part B, chapter 4) reports the synthesis of a self-fabricated co-polymer for selective isolation of caffeine from black and green tea samples. Therefore, glycidyl methacrylate and ethylene glycol dimethacrylate were polymerized and further modified with histidine. For investigation of maximum adsorption capacity and adsorption kinetics several batch adsorption experiments were executed. The synthesized sorbent showed excellent selectivity for the isolation of caffeine from tea samples in comparison to commercially available sorbent types.<br />The fourth publication (part B, chapter 5) deals with the method development of a high performance liquid chromatographic method for quantification of polycyclic aromatic hydrocarbons. Different SPE materials were checked for their performance in determination of these compounds from plant matrices. In these studies, also a self-manufactured polymer was used based on synthesis described in the second publication (part B, chapter 3). Method optimization was executed through variation of different solvents and solvent mixtures. The developed analytical method was finally evaluated according to international guidelines.<br />In the fifth publication (part B, chapter 6) novel room-temperature ionic liquids were synthesized and applied for liquid phase microextraction. After characterization using nuclear magnetic resonance spectroscopy different extraction experiments were accomplished to reveal properties of the extraction systems. Recoveries, enrichment factors as well as extraction times were investigated in aqueous solutions containing pharmaceuticals and estrogens to find optimum conditions.<br />The sixth publication (part B, chapter 7) describes the development of a molecularly imprinted polymer system for selective isolation of acetylsalicylic acid from aqueous samples. Comprehensive characterization was executed and batch adsorption experiments were accomplished to determine maximum adsorption capacities, time profiles and selectivity of the material. Applicability for water treatment was checked by isolation and regeneration studies. | ||
