Sprache Salzburg, Univ., Diss., 2012 ger: Blutgerinnung ist ein effektiver Mechanismus um lebensbedrohlichen Blutverlust nach einer Gefäßschädigung zu verhindern.<br />Neben einer raschen Aktivierung, eine strenge Kontrolle ist notwendig um eine unspezifische Verbreitung des Signals und darauf folgende Thrombus Bildung zu verhindern. Um das zu bewerkstelligen entwickelte die Natur eine Reihe von Regulationsmechanismen, bestehend aus einer zweistufigen Aktivierung und starken Inhibitoren, die in Summe eine komplexe Kaskade, bestehend aus tryptischen Serin-Proteasen, ihren Kofaktoren und Inhibitoren, bilden. Wir konnten einen zusätzlichen Regulierungsmechanismus zu identifizieren. Während die Faktoren VII und IX als Einzelkette im Blutstrom zirkulieren, kommt deren Homolog Faktor X als Doppelkette vor.<br />Unsere Daten zeigten, dass diese pre-Prozessierung zu einer Entspannung innerhalb des Aktivierungs-Peptids führt welches daraufhin von physiologischen Bindepartnern schlechter erkannt wird. Zusätzlich konnten wir zeigen, dass diese konformationelle Entspannung direkt mit einer neu identifizierten Liganden Bindestelle vernetzt ist. Diese Bindestelle kann als mögliches Ziel für die Entwicklung therapeutischer Substanzen dienen.<br />Ein neuer Ansatz um die Symptome von Hämophilie zu mindern ist die Verlängerung des Gerinnungssignals über den extrinsischen Weg. Um das zu bewerkstelligen hat man in den letzten Jahren Substanzen entwickelt die effektiv tissue factor pathway inhibitor (TFPI) inhibieren. Wir konnten die Kristall-Strukturen von TFPI komplexiert mit inhibitorischen Peptiden lösen. Dadurch bekamen wir nicht nur einen Einblick in ihren Wirkmechanismus sondern konnten sie auch weiter optimieren.<br />Urokinase und fXa sind beide tryptische Serin-Proteasen die in entgegen gesetzten Wegen der Gerinnungsbalance vorkommen. Da sie dieselbe Anordnung der katalytischen Triade und auch der Substrat bindenden S1 Tasche zeigen, testeten wir die Kreuz-Reaktivität von Urokinase spezifischen Inhibitoren auf fXa. Weiters spezifizierten wir die inhibitorischen Eigenschaften und die Kreuz-Reaktivität einer neuen Klasse von niedrig molekularen fXa Inhibitoren mittels kinetischer Analysen.<br /> eng: Blood coagulation is an effective mechanism to prevent life threatening blood loss after vascular injury. Beside a rapid onset of the signal, a strict down-regulation is indispensable to avoid spreading of the signal and spontaneous thrombus formation. To ensure this prerequisites nature evolved a series of tight regulation mechanisms, including a two step activation mechanism and potent inhibitors, resulting in a complex cascade of trypsin-like serine proteases, their cofactors and inhibitors. However, we were able to identify an additional break, dampening spontaneous clot formation. Whilst coagulation factors VII and IX appear as single chain forms within the blood stream, their homologue factor X circulates as a pre-processed two chain form. We found that the pre-processing leads to a conformational disorder of the activation peptide influencing the activation behavior of factor X by endogenous and exogenous activators. We also found that the conformational state of the activation peptide is strictly linked to a high affinity benzamidine/arginine binding site which offers novel opportunities to develop therapeutic compounds for bleeding disorders.<br />A new approach to counteract the symptoms of hemophilia is prolonging the signal of the extrinsic pathway. Thus in recent years efforts were being made to find and optimize synthetic compounds inhibiting tissue factor pathway inhibitor (TFPI). We solved the crystal structure of TFPI in complex with inhibitory peptides which revealed not only their mode of action but also enabled us to make a structure based sequence optimization of the inhibitors. Urokinase and fXa are both trypsin-like serine proteases which act in opposing pathways of blood clot balance. Whilst fXa leads to clot formation, urokinase plays an important role in thrombolysis. As they share the same arrangement of the catalytic triad as well as the same selectivity in the S1 pocket we analyzed the cross-reactivity behavior of urokinase specific, active site directed compounds on fXa.<br />Furthermore we kinetically specified the inhibition properties and cross-reactivity behaviour of a new class of small molecular weight fXa inhibitors.
