Die Freisetzung von HCN dient vermutlich als Abwehrmechanismus gegen Pflanzenfresser. Die Rückreaktion ist relevant für industrielle Biokatalyse.<br />Die Hydroxynitrillyase aus Linum usitatissimum (LuHNL) zeigt (R)-Enantioselektivität und hat Acetoncyanhydrin und Butanoncyanhydrin als biologische Substrate. Das Enzym zeigt signifikante Homologie zu zinkabhängigen Alkoholdehydrogenasen. Das Ziel dieser Arbeit war die Reinigung, Kristallisation und Bestimmung der nativen 3D Struktur der LuHNL und die Aufklärung des katalytischen Mechanismus.<br />Das kommerziell erhältliche Enzym wurde mittels hydrophober Austauschchromatographie, Größenausschlusschromatographie und präparativer Gelelektrophorese gereinigt. Nach der Kristallisation mit der hanging drop Technik wurden Streudaten gesammelt. Die Tatsache, dass LuHNL Zink enthält, ermöglichte die Strukturlösung mittels multipler anomaler Streuung. Am Ende der Verfeinerung betrug der kristallographische R-Wert 22,0% (freier R-Wert = 25,8%) bei einer Auflösung von 2,3 Å.<br />LuHNL enthält einen NAD+-Kofaktor, zwei Zinkionen und ein Natriumion pro Monomer von 422 Aminosäuren. Die Faltung ist sehr ähnlich wie bei zinkabhängigen Alkoholdehydrogenasen. Jedes LuHNL Monomer besteht aus zwei Domänen die durch eine Spalte, in der auch das aktive Zentrum liegt, getrennt sind. Die kofaktorbindende Domäne besitzt einen Rossman Fold und ist für die Dimerisierung des Proteins verantwortlich. Die substratbindende Domäne besitzt eine alpha/beta Struktur und bindet die zwei Zink- und das Natriumion.<br />Das aktive Zentrum liegt neben dem katalytisch aktiven Zinkion und dem NAD+ Kofaktor. Das Zinkion wird von zwei Cysteinen, einem Histidin und einem Wassermolekül koordiniert. (R)-Butanon wurde mit dem Programm Autodock ins aktive Zentrum gedockt, was zwei unterschiedliche Substratbindungsmoden ergab. At the close of the refinement the crystallographic residuals were R = 22,0% and Rfree = 25.8% at a resolution of 2.3 Å.<br />LuHNL contains one NAD-cofactor, two zinc ions and one sodium ion per monomer of 422 amino acids. The overall fold is very similar to that of zinc dependent alcohol dehydrogenases. Each LuHNL monomer consists of two domains separated by the active site cleft. The cofactor binding domain has a Rossman Fold and binds the NAD+ cofactor. It is also responsible for the dimerisation of the protein.<br />The substrate binding domain has a/b structure and a GroEs like fold and binds the two zinc and a sodium ion.<br />The active site of LuHNL lies next to the catalytically active zinc ion and the bound NAD+ cofactor. The zinc ion is coordinated by two cysteins and a histidine and a water molecule. (R)-butanone cyanohydrin was docked into the active site, which yielded two different reasonable substrate binding modes.
