Im Vergleich zum vollstaendigen, 11 Exons umfassenden SCGN Genprodukt Secretagogin, sind in der Variante Exon 2 und Exon 7 herausgespliced, was eine Verschiebung des Leserahmens ab der 28. Aminosaeure zur Folge hat. Dies fuehrt zu einem Stopcodon nach der 49. Aminosaeure. Das Protein, namens Setagin, besitzt im Vergleich zum Schwestergenprodukt Secretagogin kein kalziumbindendes EF-Hand-Motiv. Die rekombinante Herstellung wurde in E.coli als GST-Fusionsprotein durchgefuehrt. Es konnten Setagin-spezifische monoklonale Antikoerper erzeugt werden, anhand derer der Nachweis des endogenen Peptides im Pankreas (Langerhans-Inseln), wie auch in verschiedenen Regionen des Gehirns immunologisch erfolgte. Eine Proteinfunktionsanalyse wurde mittels Insulinpromotor-Luziferase-Reporterassays durchgeführt. Es konnte dabei ein inhibitorischer Effekt des Setagin auf die Insulintranskription aufgezeigt werden. Des Weiteren wurden im Rahmen dieser Arbeit in Tumoren Proteinexpressionsanalysen durchgeführt, um für neuroendokrine Neoplasien spezifische Proteinexpressionsmuster zu identifizieren. Mittels Immunisierung von Balb-C Mäusen mit humanem Insulinomgewebsextrakt wurden monoklonale Antikoerper produziert, von denen einer (1D4) ein in neuroendokrinen Tumoren ueberexprimiertes Protein erkannte. Immunhistochemisch wurde das Antigen in einzelnen Zellen der Adenohypophyse und in hypothalamischen Neuronen nachgewiesen. Das Antigen des monoklonalen Antikoerpers 1D4 konnte nach Aufreinigung massenspektrometrisch der Hsp70-Proteinfamilie zugeordnet werden. Mittels des monoklonalen 1D4 Antikoerpers wurden unterschiedliche Hsp70-Isoformen im Hypothalamus und Insulinomgewebe nachgewiesen. Ferner war eine tumorspezifische trunkierte Hsp70-8 Variante in den untersuchten neuroendokrinen Tumoren aufgefallen. eng: In this project with the main focus on neuroendocrine tumors a novel splice variant of the SCGN gene, which is located at chromosome 6, has been identified by RT-PCR and Northern analysis. The C-terminally truncated product, named SETAGIN, is homologous at the 27 N-terminal amino acids (aa) with its already known sister gene product secretagogin, a calcium binding protein. Because of the alternative splicing and consecutive frame shift the primary structure of the peptide is changed in the C-terminal region and it has no putative calcium binding site. The 49 aa long peptide could be produced by recombinant gene techniques. Using antibodies, generated against the SETAGIN-specific part of the peptide, the native peptide could be visualized in the pancreatic islets of Langerhans and cerebellum by immunocytochemistry as well as by immunoblot, and further in hypothalamus and pituitary. The biological function of SETAGIN has been determined, as an endogenous dominant negative against secretagogin when insulin transcription is measured by insulin promoter driven luciferase reporter assays. To identify neuroendocrine tumor specific protein expression, mAbs with a tumor specific reaction pattern were generated using a human insulinoma as immunogen. One of these mAbs (1D4 mAb) showed striking immunoreactivity against various neuroendocrine tumors without staining pancreatic islets of Langerhans. Furthermore, 1D4 mAb immunostained a characteristic subtype of hypothalamic neurons and infrequently cells in the anterior part of the pituitary.
