Um den Einfluss von geometrischen Faktoren auf die Zellphysiologie zu studieren, wurde eine Methode zur Modifikation von Polymeroberflächen entwickelt, die es erlaubt, die Zellform und -größe genau zu kontrollieren. Dazu wurde in einem photolithographischen 'lift-off' Verfahren ein Muster aus einem protein- und zelladhäsiven Fluorocarbon (FC)-Polymer auf einem adhäsionsresistenten Hintergrund aufgebracht. Dieser wurde durch eine Plasma-Polymerisation von Tetraethylenglykoldimethylether (Tetraglyme) hergestellt. Die Topographie und Muster-Qualität sowie die molekulare Struktur dieser Oberflächen wurden mittels Rasterkraftmikroskopie, Röntgen-Photoelektronenspektroskopie (XPS) und Sekundärionenspektroskopie (SIMS) untersucht. Die Proben zeigten eine hohe geometrische Detailtreue sowie die erwünschte Polymerverteilung. Glatte Muskelzellen einer unsterblichen Zellinie (FR 344), auf diese gemusterten Substrate ausgebracht, verblieben auf dem Muster, ohne sich in die benachbarten Regionen auszudehnen. Diese Größen- und Geometriekontrolle kann über mindestens zwei Wochen ohne eine Ablösen oder Absterben der Zellen aufrechterhalten werden. Weiters wurde eine Möglichkeit erarbeitet, diese Muster mit spezifischen Zellaffinitätssignalen zu ergänzen. Dazu wurde ein Adhäsionspeptid (YIGSR) kovalent an den Alkoholterminus eines oberflächenaktiven FC-Polyethylenglykol Moleküls gebunden. Die Adsorption an plasmapolymerisierte Fluorocarbon-Filme wurde mittels SPR (surface plasmon resonance) untersucht. Es konnte mittels SIMS gezeigt werden, dass sich dieses Molekül aus wässriger Lösung nur am hydrophoben FC-Muster anlagert, während der Hintergrund unmodifiziert blieb. eng: This thesis describes the development of a novel surface modification technique to manufacture micro-patterned cell culture substrates for the control of the cell shape and projected area of smooth muscle cells. A micrometer size pattern of a thin fluorocarbon polymer is deposited on a transparent polymeric substrate (PET) that had been rendered non-fouling by the deposition of a tetraglyme polymer. The topography and chemical composition was analyzed using atomic force microscopy, electron spectroscopy for chemical analysis (ESCA) and secondary ion mass spectroscopy (SIMS) in the imaging mode. The substrates showed excellent pattern fidelity and the desired chemical composition. It was demonstrated that the non-fouling background polymer resisted the adsorption of proteins and cells, while the fluorocarbon pattern showed a high affinity for those components. Smooth muscle cells from an immortal cell line (FR 344), for which the potential for redifferentiation had been demonstrated, were seeded onto these patterned cell culture substrates. After incubation of up to 14 days the cells were analyzed using fluorescence microscopy, showing that cells remained vital and control over the shape and size of the cells was achieved and maintained for the duration of the experiments. Furthermore a method to modify the patterned substrates with specific affinity ligands was developed. A cell-adhesion peptide derived from laminin was conjugated to a FC-PEG surfactant. The binding of such a peptide-surfactant conjugate with histamine as a model ligand to plasma-deposited FC polymers was studied using surface plasmon resonance.
